Strep-tag® technology
Cutting-edge tools for high-performance research in cell and protein isolation are enabled by the Strep-tag® technology, leveraging the robust interaction between biotin and streptavidin.
This technology harnesses the specificity and controllability of the interaction between the synthetic Strep-tag®II peptide and engineered streptavidins like Strep-Tactin®, renowned for its stability. By occupying the biotin binding site, Strep-tag®II forms a reversible interaction with engineered streptavidins, Strep-Tactin® and Strep-Tactin®XT easily disrupted by excess biotin.
The Strep-tag®II, comprising eight amino acids (Trp-Ser-His-Pro-Gln-Phe-Glu-Lys), and its variant, Twin-Strep-tag® is composed of 28 amino acids inluding two of these motifs in tandem connected by a linker. Both exhibit varying affinities for Strep-Tactin® derivatives. Strep-tag®II shows significantly higher affinity for Strep-Tactin® compared to streptavidin, with an affinity nearly 100 times stronger. The development of Strep-Tactin®XT further enhances affinity, which shares nanomolar affinity with Strep-tag®II and picomolar affinity for Twin-Strep-tag®.
Due to variations in binding strength among different tag-ligand combinations, the Strep-tag® system has emerged as a versatile tool for isolating proteins and cells.
The Strep-tag® fused to recombinant proteins facilitates efficient one-step purification when immobilized on Strep-Tactin®.
When attached to antibody-derived Fab fragments or nanobodies, the Twin-Strep-tag® interacts with multimerized Strep-Tactin®, enabling the capture and release of target cells based on their surface markers or antigen specificity.
Strep-Tactin® (or Strep-Tactin®XT) can be conjugated to microplates, fluorophores, or chips, wich makes this tool suitable for a wide range of analytical applications following the isolation of target material, such as detection, immobilization, and interaction studies.
