The adeno-associated virus (AAV) is a small, non-enveloped virus and have a single-stranded DNA (ssDNA) genome of approximately 4.7 kb. Twelve different AAV serotypes are known.
The AAV genome is flanked by two terminal repeats (ITRs) essential for packaging the viral DNA, replication, transcription and site-specific integration. The genome consist of the Rep gene that encodes four rep proteins which are essential for DNA replication, packaging the viral genome and its integration into the host DNA. Also the Cap gene is present in its genome which encodes three viral capsid proteins (VP) that form the virus capsid.
AAV is interested to several gene therapy researcher due to several features such as lack of pathogenicity, the ability to also infect non-dividing cells and has the ability to stably integrate into the host genome at a specific site.
AAV derived vectors possesses very low cytotoxicity and low immunogenicity and rarely integrate into the genome but reside as episomes in the nucleus, lowering the chance of spontaneous gene alteration. AAV enables long-term transgene expression so in theory genes only need to be delivered once. These features makes the AAV-derived vectors a perfect vehicle for the delivery of genetic material to human cells.

Antibodies
The antibodies are great tools to characterize the different stages of AAV infection and to analyze the AAV assembly process. The AAV capsid protein antibodies recognize the linear epitopes of the viral capids, so specifically reactis with denaturated AAV capsid proteins. The AAV particle antibodies recognize a conformational epitope of the assembled capsids, so specifically react with intact AAV particles. Also AAV replicase antibodies are available for your research.

ELISAs

Testing individual and crude AAV samples quick during development and manufacturing is often difficult, because most accurate analytical AAV tools are not convenient for single sample measurement and/or require highly purified samples. Optiprep™ solution
Optiprep™ Cat.no 1893 is the ideal solution for virus purification such as AAV, because the ratio of infectious particles to total particles is 100 times higher using Optiprep™ gradients compared to CsCl gradients.
Using OptiPrepTM for AAV purification saves time, because no need to remove iodixanol for most downstream processes, it causes little or no damage to viral particles, results in a then times greater recovery rate than CsCl gradient alternatives and results in a higher infectively titer with no toxic effects.
