Bcl-2-associated anthanogene 1 (BAG-1) is a multifunctional co-chaperone protein with roles in apoptosis and transcription, as well as cell motility, survival, and proliferation.{58201,58200} It is encoded by a single gene, BAG1, that produces several isoforms with variable N-termini through alternate translation initiation of a single mRNA transcript.{58202} BAG-1L, the longest isoform, is localized to the nucleus and composed of a nuclear localization sequence, a basic DNA-binding domain, a ubiquitin-like domain, and an Hsp70- and Hsc70-binding domain.{58200} It functions as a co-chaperone to enhance transactivation of the androgen receptor via interaction with the partially folded and intrinsically disordered N-terminal tau-5 domain, which stabilizes the receptor to facilitate chromatin binding and activates gene transcription.{58203} Overexpression of BAG-1L inhibits apoptosis induced by cisplatin (Item No. 13119) in HeLa cells stably transfected with pEZ-M02-BAG-1L plasmid DNA, and BAG-1L levels are increased in tumors isolated from patients with castration-resistant prostate cancer (CRPC).{58202,58203} Cayman’s BAG-1L Rabbit Monoclonal Antibody can be used for immunocytochemistry (ICC), immunohistochemistry (ICC), and Western blot (WB) applications.
